目的:探讨奥曲肽对肝癌细胞SMMC-7721转化生长因子-α(TGF-α)自分泌及对其促进细胞增殖的影响。方法:运用放射免疫法和逆转录-聚合酶链反应(RT-PCR)检测TGF-α的表达,运用免疫组化法和RT-PCR检测表皮生长因子受体(EGFR)的表达,细胞增殖变化以流式细胞术和集落形成实验检测。结果:加入奥曲肽后,上清液中TGF-α含量在相应时间点均较对照组明显减少,抑制率为15.0%~26.7%;TGF-αmRNA指数较对照组降低。TGF-α使EGFRmRNA及蛋白表达增加,而奥曲肽则抑制它们的表达。奥曲肽和TGF-α同时作用于细胞时,细胞增殖指数(PI)值和集落形成率比TGF-α单独作用时明显降低。结论:人肝癌细胞系SMMC-7721中存在TGF-α自分泌环路,并促进该细胞增殖,奥曲肽能抑制此细胞系TGF-α自分泌,从而抑制细胞增殖。
Objective: To investigate the effect of octreotide on the autocrine of transforming growth factor-α (TGF-α) and its proliferation in hepatocellular carcinoma cell line SMMC-7721. Methods: The expression of TGF-α was detected by radioimmunoassay and reverse transcription-polymerase chain reaction (RT-PCR). The expression of epidermal growth factor receptor (EGFR) was detected by immunohistochemistry and RT-PCR. Flow cytometry and colony formation assay. Results: After addition of octreotide, the content of TGF-α in the supernatant decreased significantly at the corresponding time points compared with the control group, with the inhibition rate of 15.0% -26.7%. The TGF-αmRNA index was lower than that of the control group. TGF-α increases EGFR mRNA and protein expression, whereas octreotide inhibits their expression. When octreotide and TGF-α were simultaneously administered to cells, the cell proliferation index (PI) and colony formation rate were significantly lower than those of TGF-α alone. CONCLUSION: TGF-|Á autocrine loop exists in human hepatocellular carcinoma cell line SMMC-7721 and promotes cell proliferation. Octreotide can inhibit the autocrine secretion of TGF-|Á and suppress the proliferation of the cell line.
