目的测定自主建立的转基因致癌性模型C57-ras小鼠与BALB/c小鼠杂交1代的生长发育特性参数,为该模型的商业化供应提供基础数据。方法将雄性转基因阳性C57-ras小鼠与野生型雌性BALB/c小鼠交配,获得杂交1代(CB6F1 c-Ha-ras+/-)并测定平均产仔数、离乳率、阳性鼠生长发育情况、转基因阳性率等参数。结果在生长发育方面,雌鼠平均每窝产子数为8只,离乳率为90%,雄雌性别比符合预期。CB6F1的平均出生重为(1.73±0.05)g,10周龄时阳性鼠平均体重雌性(24.38±1.74)g,雄性(29.42±1.72)g。雄性鼠体重明显高于雌性,差异存在极显著性(P<0.01);基因型检测阳性率为46.9%,符合遗传规律。结论转基因致癌性模型C57-ras小鼠与BALB/c小鼠杂交1代的生长发育特性正常,可实现规模化商业供应。
OBJECTIVE: To determine the growth and development parameters of the first generation of C57-ras transgenic mice and BALB / c mice established by ourselves, providing the basic data for the commercialization of the model. Methods Male transgenic C57-ras mice were mated with wild-type female BALB / c mice to obtain the first generation of hybrids (CB6F1 c-Ha-ras +/-) and the average number of litter size, Situation, gene positive rate and other parameters. Results In the aspect of growth and development, the average number of female births per litter was 8 and the rate of milk release was 90%. The male-female ratio was in line with the expectation. The average birth weight of CB6F1 was (1.73 ± 0.05) g and the average body weight of female mice (24.38 ± 1.74g) and male (29.42 ± 1.72g) were positive at 10 weeks of age. The body weight of male rats was significantly higher than that of females, the difference was significant (P <0.01). The positive rate of genotype was 46.9%, which accorded with the genetic rule. Conclusion The growth and development of transgenic C57-ras mice and BALB / c mice are normal and the large-scale commercial supply can be achieved.
