目的:探讨人肺腺癌A549细胞系SP细胞亚群体外培养的生物学行为特征。方法:免疫组化方法检测A549细胞系AB-CG2蛋白的表达;流式细胞仪分选A549细胞系SP细胞和非SP细胞亚群,用RT-PCR方法检测两亚群细胞ABCG2基因的表达;并通过两亚群细胞的生长曲线、细胞分裂指数、细胞周期各时相分布以及平板克隆形成实验、迁移和侵袭研究两亚群细胞的生物学行为。结果:A549细胞系ABCG2蛋白表达的阳性率约为2%;SP细胞表达ABCG2,而非SP细胞则不表达;两亚群细胞的增殖和迁移能力相似(P>0.05);SP细胞的克隆形成率和侵袭能力则大于非SP细胞(P<0.05)。结论:A549细胞系SP细胞亚群体外成瘤能力及侵袭能力强于非SP细胞亚群,SP细胞亚群可能富集了肿瘤干细胞。
Objective: To investigate the biological behavior of human lung adenocarcinoma A549 cell line SP cells in vitro culture. Methods: The expression of AB-CG2 protein in A549 cell line was detected by immunohistochemistry. The SP cells and non-SP cell subsets of A549 cell line were sorted by flow cytometry. The expression of ABCG2 gene in two subpopulations was detected by RT-PCR. The biological behavior of the two subpopulations was studied by cell growth curve, cell division index, cell cycle distribution, plate clone formation assay, migration and invasion. Results: The positive rate of ABCG2 protein in A549 cell line was about 2%. The expression of ABCG2 in SP cells was not expressed in SP cells. The proliferation and migration of two subpopulations were similar (P> 0.05). The clonogenicity of SP cells Rate and invasive ability was greater than non-SP cells (P <0.05). CONCLUSION: The ability of SP cell subsets of A549 cells to grow in vitro is stronger than that of non-SP cells in vitro. The SP cells may be enriched in tumor stem cells.
