背景:急性心肌缺血时c-JunN末端蛋白激酶(c-JunN-terminalproteinkinase,JNK)被激活,并使得缺血损伤加重。慢性冬眠心肌组织中JNK的亚型———JNK1/2是否被激活及其在慢性冬眠心肌发生发展机制中的作用又是什么呢?目的:明确慢性冬眠心肌组织中JNK1/2的蛋白表达及其磷酸化(p-JNK1/2)的变化。设计:随机对照的实验研究。单位:徐州医学院附属医院心血管病研究所。材料和方法:在徐州医学院附属医院导管室进行动物模型的制备、在徐州医学院生化教研室测定JNK1/2的蛋白表达及其磷酸化(p-JNK1/2)的变化。将14只小型中国家猪随机分为实验组(n=8)与对照组(n=6)。实验组以右冠状动脉为靶血管,经右股动脉送入自制的缩窄器,制备成慢性冬眠心肌及心肌梗死的模型。获取对照组心肌组织、实验组中的正常心肌组织及慢性冬眠心肌组织的样本进行光镜、电镜检查并采用免疫印迹(Westernblotting)分析3组心肌组织的JNK1/2的蛋白表达及其磷酸化的变化。主要观察指标:慢性冬眠心肌组织中JNK1/2是否被活化。结果:实验组慢性冬眠心肌组织p-JNK1/2比实验组正常心肌组织、对照组心肌组织高。对照组、实验组正常心肌组织、实验组慢性冬眠心肌组织p-JNK1/2的免疫活性分别为1,1.42±0.52,2.6±0.59。结论:慢性冬眠心肌组织中JNK1/2被激活
BACKGROUND: C-Jun N-terminal protein kinase (JNK) is activated during acute myocardial ischemia and aggravates ischemic damage. JNK subtypes in chronic hibernating myocardium --- JNK1 / 2 is activated and its role in the pathogenesis of chronic hibernating myocardium What is the purpose? To determine the chronic hibernating myocardium JNK1 / 2 protein expression and Its phosphorylation (p-JNK1 / 2) changes. Design: Randomized controlled experimental study. Unit: Xuzhou Medical College Hospital Cardiovascular Institute. MATERIALS AND METHODS: The animal model was established in the catheterization room of Affiliated Hospital of Xuzhou Medical College. The expression of JNK1 / 2 protein and the change of its phosphorylation (p-JNK1 / 2) were determined in the Department of Biochemistry, Xuzhou Medical College. Fourteen small Chinese domestic pigs were randomly divided into experimental group (n = 8) and control group (n = 6). Experimental group, the right coronary artery as the target vessel, the right femoral artery into the homemade constriction, preparation of chronic hibernating myocardium and myocardial infarction model. The samples from control group, normal myocardium and chronic hibernating myocardium were detected by light microscope and electron microscope. The protein expression of JNK1 / 2 and the phosphorylation of JNK1 / 2 in myocardium were detected by Western blotting Variety. MAIN OUTCOME MEASURES: Is JNK1 / 2 activated in chronic hibernating myocardium? Results: The p-JNK1 / 2 of chronic hibernating myocardium in the experimental group was higher than that in the normal myocardium and the control group in the experimental group. The immunocompetence of p-JNK1 / 2 in control group, experimental myocardial tissue and experimental hibernating myocardium were 1, 1.42 ± 0.52 and 2.6 ± 0.59, respectively. Conclusion: JNK1 / 2 is activated in chronic hibernating myocardium
