目的研究大鼠灌胃给予当归补血汤后阿魏酸在体内的组织分布特性。方法采用高效液相色谱法;色谱柱为Kro-masil C18(4.6mm×150mm,5μm);流动相为0.2%甲醇乙腈-0.2%磷酸0.2%三乙胺水溶液(18:82);检测波长322nm;柱温40℃;流速0.8ml/min。结果各组织中的阿魏酸在10~400ng范围内与峰面积呈良好的线性关系;回收率平均为87.97%±1.95%;日内和日间的RSD分别1.92%~3.87%和3.21%~6.39%之间。阿魏酸在各组织器官中浓度差异较大。结论大鼠灌胃给予当归补血汤后阿魏酸主要在肝脏和肾脏分布,阿魏酸不能通过血脑屏障。
Objective To study the tissue distribution of ferulic acid in rats after intragastric administration of Danggui Buxue Decoction. The method was performed by high performance liquid chromatography. The chromatographic column was Kro-masil C18(4.6mm×150mm, 5μm). The mobile phase was 0.2% methanol acetonitrile-0.2% phosphoric acid 0.2% triethylamine aqueous solution (18:82). The detection wavelength was 322nm. Column temperature 40°C; flow rate 0.8 ml/min. Results The ferulic acid in each tissue showed a good linear relationship with the peak area in the range of 10-400 ng; the average recovery was 87.97%±1.95%; the intraday and inter-day RSD were 1.92%-3.87% and 3.21%-6.39, respectively. %between. The concentration of ferulic acid in various tissues and organs varies widely. Conclusion Ferulic acid is mainly distributed in the liver and kidney after intragastric administration of Dangguibuxue decoction. Ferulic acid cannot pass through the blood-brain barrier.
