目的:研究伤寒沙门菌非编码RNA Asr C对其在巨噬细胞中生存能力的影响及机制。方法:用Asr C高表达菌株和空质粒对照株分别感染巨噬细胞12,24 h,检测伤寒沙门菌在感染细胞中的生存能力;qRT-PCR检测asr C、rse C、rpo E mRNA的表达水平;蛋白质印迹检测自噬标志蛋白LC3的表达。结果:与空质粒对照株相比,Asr C高表达菌株感染巨噬细胞12,24 h后在巨噬细胞中的生存能力明显下降(P均<0.01),asr C mRNA表达水平增加(P均<0.05),对侧靶基因rse C mRNA的表达水平增高(P均<0.05),而rpo E mRNA的表达水平无明显变化(P>0.05);Asr C高表达菌株自噬标志蛋白LC3的表达与空质粒对照株间的差异无统计学意义(P>0.05)。结论:伤寒沙门菌非编码RNA Asr C高表达可降低其在巨噬细胞中的生存能力,可能与自噬无直接关系。
Objective: To study the effect and mechanism of non-coding RNA Asr C on the viability of macrophages in Salmonella typhi. Methods: The macrophages were infected with Asr C high expression and empty plasmid control strains respectively for 12,24 h to detect the viability of Salmonella typhimurium in infected cells. The expression of asr C, rse C and rpo E mRNA was detected by qRT-PCR Western blotting was used to detect the expression of autophagy marker LC3. RESULTS: Compared with the control plasmid, the viability of macrophages in macrophages infected with Asr C overexpression strain was significantly decreased (P <0.01) and the expression of asr C mRNA increased (P (P <0.05), while the mRNA level of srse C mRNA increased (P <0.05), while the mRNA expression of rpo E had no significant change (P> 0.05). The expression of autophagy marker LC3 There was no significant difference between the control group and empty plasmid (P> 0.05). Conclusion: High expression of Asn C RNA can reduce its viability in macrophages and may not be directly related to autophagy.
