应用随机引物扩增片段多态性 (RAPD)技术对 2种紫菜的 5个品系进行遗传多样性分析。共筛选出 2 1条随机引物 ,PCR反应得到 147条扩增片段。根据共享扩增片段计算遗传相似性指数 (F)和相对遗传距离 ,利用NJ法构建系统树。结果表明 ,条斑紫菜或坛紫菜的养殖品系首先聚类在一起 ,两个条斑紫菜养殖品系之间的遗传距离是0 32 ,两个坛紫菜养殖品系之间的遗传距离是 0 .31。条斑紫菜养殖品系CPY1 0 8A与坛紫菜养殖品系CPH8 83之间的遗传距离最大 ,达 0 .4 2。本文结果提示RAPD可以作为简便有效的分子工具应用于紫菜的遗传多样性和种质鉴定研究中。
Random amplified polymorphic DNA (RAPD) technique was used to analyze the genetic diversity of five species of two laver cultivars. A total of 21 random primers were screened and 147 amplified fragments were obtained by PCR. Based on the shared amplified fragments, genetic similarity index (F) and relative genetic distance were calculated, and the phylogenetic tree was constructed by NJ method. The results showed that the cultured strains of Porphyra yezoensis or Porphyra yezoensis were first clustered together, the genetic distance between the two Porphyra culture cultivars was 0 32, and the genetic distance between the two cultivated Porphyra culture cultivars was 0.31. The genetic distance between P. yunnaneus cultivar CPY1 0 8A and algae laver cultivar CPH8 83 was the largest, reaching 0.42. Our results suggest that RAPD can be used as a simple and effective molecular tool in genetic diversity and germplasm identification of seaweed.
