[目的]用B交配型位点基因对杏鲍菇不同极性菌株进行快速鉴定。[方法]根据杏鲍菇B交配型位点基因的信息素PEphb(3.3.1~3.3.4)和信息素受体PEPHSTE(3.3.1~3.3.4)基因设计8对特异性引物,对原生质体单核化得到的2个不同极性的杏鲍菇样本进行PCR扩增。[结果]4个信息素基因和3个信息素受体基因仅能在A2B2极性中存在,而在A1B1极性中不存在。[结论]用B交配型位点基因能对杏鲍菇不同极性进行鉴定,且这7个基因可以作为杏鲍菇原生质体单核化极性快速鉴定的SCAR分子标记。
[Objective] The rapid identification of different polarity strains of Pleurotus eryngii with B mating type locus genes. [Method] Eight pairs of specific primers were designed according to the genes pheromone PEphb (3.3.1 ~ 3.3.4) and pheromone receptor PEPHSTE (3.3.1 ~ 3.3.4) of B mating type locus, Two samples of Pleurotus eryngii with different polarities obtained from protoplast nucleation were amplified by PCR. [Result] The four pheromone genes and three pheromone receptor genes could only exist in A2B2 polarity, but not in A1B1 polarity. [Conclusion] The different polarity of Pleurotus eryngii can be identified by using the B mating type locus genes, and these 7 genes can be used as SCAR molecular markers for rapid identification of protoplast mononuclear polarity in Pleurotus eryngii.
