目的分离和鉴定猪链球菌,为诊断和治疗提供科学依据。方法用无选择性的血琼脂平板对患者血液、脑脊液、尸检组织和病死猪内脏进行分离培养,并对分离到的细菌进行染色形态观察;用PCR技术检测猪链球菌种基因、荚膜多糖编码基因、溶菌酶释放相关蛋白基因和溶血素基因;用VITEK2compact微生物生化鉴定仪定型。结果从4例患者血液、2例患者脑脊液、2例患者尸检组织和7例病死猪中分离到27株链球菌,经鉴定27株菌均为猪链球菌2型,猪链球菌种基因和猪链球菌三种毒力基因阳性。结论此次疫情为猪链球菌2型引起。
Objective To isolate and identify Streptococcus suis, to provide a scientific basis for diagnosis and treatment. Methods The blood, cerebrospinal fluid (CSF), autopsy tissues and diseased pig offal were isolated and cultured with nonselective blood agar plates. The stained morphology of the isolated bacteria was observed. The gene encoding Streptococcus suis spp. Gene, lysozyme release related protein gene and hemolysin gene; with VITEK2compact microbial biochemical identification of stereotypes. Results 27 streptococci were isolated from blood of 4 patients, cerebrospinal fluid of 2 patients, autopsy tissue from 2 patients and 7 dead pigs. All 27 strains were identified as Streptococcus suis type 2, Streptococcus suis type and pig Streptococcus three virulence gene positive. Conclusion The outbreak was caused by Streptococcus suis type 2.
