目的:采用鸭乙型肝炎动物模型考核复方金牡蛎胶囊的抗病毒作用和观察药物毒性等作用。方法:实验动物为垂直传播感染、DHBVDNA检测阳性麻鸭,实验分设0.2,0.6和1.8g/kg的不同剂量组和3个分别为DHBVDNA(+)、DHBVDNA(-)和阿昔洛韦阳性药物对照组。给药为口腔途径喂饲,每周3次隔日给药,连续给药12周。结果:不同剂量组血清DHBVDNA平均含量在用药后均有不同程度下降,在停药后第2周时仍基本维持于停药时的水平。3个月的用药期间,各组分别有4~7只麻鸭血清DHBVDNA转为阴性,其中高剂量组的转阴率达到58%。病理观察有一定的病理改善作用,减轻炎症程度,尤以中、低剂量组的为明显,未发现药物有明显的肝脏病理毒性作用。结论:复方金牡蛎胶囊影响鸭乙肝病毒DNA的复制,降低血清DHBVDNA含量,有较明显的抗病毒作用。
OBJECTIVE: To evaluate the antiviral effects of compound Jinyuyu capsules and to observe the effects of drug toxicity by using a duck/hepatitis B animal model. METHODS: The experimental animals were vertically transmitted infections and DHBV DNA-positive ducks. The experiments were divided into different dosage groups of 0.2, 0.6 and 1.8 g/kg, and three were DHBV DNA (+), DHBV DNA (-) and A. Ciprovir-positive drug control group. The administration was oral route feeding, 3 times a week, every other day for 12 weeks. Results: The average serum DHBV DNA levels in different dose groups decreased to varying degrees after administration, and remained at the level of withdrawal at 2 weeks after drug withdrawal. During the 3 months of drug use, 4 to 7 ducks had DHBV DNA negative in each group, and the high-dose group had a negative conversion rate of 58%. Pathological observation had some pathological improvement and reduced inflammation, especially in the medium and low dose groups. There was no obvious liver pathological toxicity. Conclusion: Compound Jinyinyu Capsules can affect DNA replication of Duck Hepatitis B virus and reduce serum DHBV DNA content, which has obvious antiviral effect.
