In biological studies, deoxy-pentose-diphenylamine reaction is a commonly used method for the quantitative determination of DNA content. However, this reaction is often less sensitive if the DNA concentration in the solution is less than 5 micrograms per milliliter. It has been reported that if the blue deoxy pentose-diphenylamine complex formed from the reaction is extracted from the solution with amyl acetate, the sensitivity can be increased four-fold and the concentration in the solution can be accurately measured as low as 1.5 Microgram of DNA.
