目的 探讨白细胞介素(IL)-4基因启动子区多态性与湖北汉族儿童哮喘的易感性及血浆IgE水平的关系。方法 用聚合酶链反应和限制片段长度多态性(PCR—RFLP)的方法对湖北汉族115例哮喘患儿及75例非哮喘对照儿童进行了+33位点多态性分析,用化学发光法测定血浆总IgE水平。结果 哮喘儿童血浆平均总IgE为351.21 IU/ml,非哮喘对照儿童血浆平均总IgE为15.48 IU/ml,有极显著性差异(P<0.0001)。哮喘患儿与非哮喘儿童IL-4基因启动子C+33T位点的等位基因频率和基因型频率CC、CT、TT之间有极显著性差异(P<0.0001),且哮喘患儿CT和TT之间均与血浆IgE升高相关(P<0.0001)。结论 IL-4启动子+33位点多态性与湖北汉族儿童哮喘易感性和血浆总IgE水平相关。
Objective To investigate the relationship between the polymorphism of interleukin (IL) -4 gene promoter and susceptibility to asthma and plasma IgE in Han children in Hubei province. Methods Polymorphism analysis of +33 loci in 115 children with asthma and 75 non-asthmatic children in Hubei Han nationality was performed by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). The chemiluminescence Total plasma IgE levels were measured. Results The average plasma IgE was 351.21 IU / ml in asthmatic children and 15.48 IU / ml in non-asthmatic children. There was a significant difference (P <0.0001). There was a significant difference (P <0.0001) between allele frequency and genotype frequency of IL-4 gene promoter C + 33T locus in asthmatic children and non-asthmatic children (P <0.0001) And TT were correlated with elevated plasma IgE (P <0.0001). Conclusion The +33 polymorphism of IL-4 promoter is associated with asthma susceptibility and total plasma IgE levels in Han children in Hubei province.
