本文研究了五种小鼠抗人大肠癌单克隆抗体:Ga_3,Aa_2,Hb_3,Cb_5,Cc-1对体外培养人大肠癌细胞株HRT-18的影响。在无补体及效应细胞存在的情况下Ca_3,Cc-1,Cb_5三种抗体能抑制靶细胞生长,呈剂量依赖型。Ca_3还可以抑制靶细胞在软琼脂中形成集落;并能使细胞膜表面超微结构发生一定改变。间接免疫荧光表明五种抗体均主要作用于细胞膜抗原,且表现出抗原表达的异质性。通过聚丙烯酰胺凝胶电泳(SDS-PAGE),电转移印迹(Western blot)结合免疫酶技术,Ga_3作用于HRT-18细胞上分子量20kd的蛋白质抗原。此外,Ga_3,Aa_2均能在人外周血单个核细胞及人补体参与下,通过ADCC及CDC效应杀伤HRT-18细胞,而不损及正常人胚肺细胞。Hb_3则只能介导CDC效应。
This study investigated the effects of five mouse anti-cancer colorectal monoclonal antibodies: Ga3, Aa2, Hb3, Cb5, and Cc-1 on human colorectal cancer cell line HRT-18 in vitro. In the absence of complement and effector cells, Ca 3, Cc-1, and Cb 5 antibodies inhibited the growth of target cells in a dose-dependent manner. Ca_3 can also inhibit target cells from forming colonies in soft agar; it can also cause certain changes in the ultrastructure of the cell membrane surface. Indirect immunofluorescence showed that all five antibodies mainly acted on cell membrane antigens and showed heterogeneity in antigen expression. By using SDS-PAGE, Western blotting and immunoenzymatic techniques, Ga 3 acted on protein antigens with a molecular weight of 20 kd on HRT-18 cells. In addition, both Ga 3 and Aa 2 can kill HRT-18 cells through ADCC and CDC effects without the damage of normal human embryonic lung cells in the presence of human peripheral blood mononuclear cells and human complement. Hb_3 can only mediate the CDC effect.
