目的采用PCR array技术探讨化瘀祛痰方对高脂血症大鼠肝脏脂质代谢信号通路相关基因mRNA表达的影响,以初步揭示化瘀祛痰方对高脂血症大鼠肝脏脂质代谢的干预作用。方法 24只健康雄性SD大鼠随机分为正常组、高脂血症组、化瘀祛痰组。高脂血症组、化瘀祛痰组予高脂饲料喂饲30天后,化瘀祛痰组予化瘀祛痰中药汤剂灌胃,正常组、高脂血症组给予等体积的生理盐水灌胃。30天后,全自动生化分析仪检测各组大鼠血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)的含量,HE染色观察肝脏形态变化,油红O染色观察肝脏脂质沉积,PCR array技术检测各组大鼠肝脏脂质代谢信号通路相关基因mRNA表达。结果与正常组相比,高脂血症组血清中TC、TG、LDL-C水平显著升高(P<0.01),HDL-C水平显著降低(P<0.01),肝细胞形成大量脂质沉积,胞浆内多出现脂肪空泡;与高脂血症组相比,化瘀祛痰组血清中TC、TG、LDL-C水平显著降低(P<0.01),HDL-C水平显著升高(P<0.05),肝脏脂滴减少。PCR array显示:化瘀祛痰组发生差异改变的基因为Abca1、Apoe、cel、Cyp39a1、Cyp46a1、Cyp7a1、Hmgcr、Ldlr、snx17。结论化瘀祛痰方可能通过调控Abca1、Apoe、cel、Cyp39a1、Cyp46a1、Cyp7a1、Hmgcr、Ldlr、snx17基因,影响胆固醇代谢,发挥其对高脂血症大鼠肝脏脂质代谢的干预作用。
Objective To investigate the effect of Huayu Qutan Recipe on mRNA expression of liver lipid metabolism signal pathway in hyperlipidemic rats by PCR array to reveal the effect of Huayu Qutan recipe on lipid metabolism in liver of hyperlipidemic rats The role of intervention. Methods Twenty-four healthy male SD rats were randomly divided into normal group, hyperlipidemia group and Huayu Qutan group. After hyperlipidemia group and Huayu Qutan group were fed with high-fat diet for 30 days, Huayu-Qutan group was treated with Huayu Qutan decoction decoction by intragastric administration. The normal group and hyperlipidemia group were given equal volume of normal saline Gavage. After 30 days, the contents of serum cholesterol, triglyceride, low density lipoprotein cholesterol and high density lipoprotein cholesterol of HDL-C were detected by automatic biochemical analyzer, Liver morphology was observed by HE staining, liver lipid deposition was observed by oil red O staining, and mRNA expression of lipid metabolism signal pathway in liver of each group was detected by PCR array. Results Compared with the normal group, the levels of TC, TG and LDL-C in serum of hyperlipidemia group were significantly increased (P <0.01), HDL-C level was significantly decreased (P <0.01) , Compared with hyperlipidemia group, the level of TC, TG and LDL-C in serum of the patients treated with Huayu Qutan was significantly lower (P <0.01) and the level of HDL-C was significantly increased P <0.05), liver lipid droplets decreased. PCR array showed that the genes that were differentially changed in Huayu Qutan group were Abca1, Apoe, cel, Cyp39a1, Cyp46a1, Cyp7a1, Hmgcr, Ldlr and snx17. Conclusion Huayu Qutan Recipe can affect the metabolism of cholesterol by regulating the expression of Abca1, Apoe, cel, Cyp39a1, Cyp46a1, Cyp7a1, Hmgcr, Ldlr and snx17 genes and exert their intervention on the liver lipid metabolism in hyperlipidemic rats.
