目的 :研究 1 92 Ir插植术治疗口腔鳞癌前后肿瘤组织 DNA含量的变化。方法 :选择 10例未经治疗的口腔癌患者 ,取放疗前新鲜肿瘤标本和放疗后 3天~ 7天手术新鲜肿瘤标本 ,通过流式细胞仪 (FCM)测定 DNA含量的 3个主要参数 ,异倍体 (aneuploid)、DNA指数 (DNA Index,DI)和 S期细胞比例 (S- phase fraction,SPF) ,比较显效组与非显效组、放疗前后 DNA含量之间的变化。结果 :显效组与非显效组的 DI分别为 1.6 3± 0 .10和 1.33± 0 .2 4(P<0 .0 5 ) ,SPF分别为 16 .6 1± 0 .86和12 .32± 4.2 0 (P<0 .0 5 ) ;放疗前后肿瘤组织 DI分别为 1.46± 0 .12和 1.2 0± 0 .11(P<0 .0 1) ,SPF分别为 14.0 3± 3.85和 8.0 8± 1.2 7(P<0 .0 1)。结论 :1 92 Ir插植术是治疗口腔鳞癌的有效方法 ,对口腔鳞癌 DNA含量的检测可以衡量放射治疗的效果并有助于制定治疗计划。
Objective: To study the changes of DNA content in tumor tissue before and after oral cavity squamous cell carcinoma treated with 1 92 Ir implantation. Methods: Ten patients with untreated oral cancer were enrolled. Fresh tumor specimens before radiotherapy and fresh tumor specimens from 3 days to 7 days after radiotherapy were collected. The three main parameters of DNA content were determined by flow cytometry (FCM) Aneuploid, DNA Index (DI) and S-phase fraction (SPF). The changes of DNA content before and after radiotherapy were compared between the two groups. Results: The DIs in the effective and non-effective groups were 1.6 3 ± 0.10 and 1.33 ± 0.24, respectively (P <0.05), the SPFs were 16.6 ± 0.86 and 12.32 ± 4.2 0 (P <0.05). The DI of tumor tissue before and after radiotherapy were 1.46 ± 0.12 and 1.2 ± 0.11 (P <0.01) respectively, the SPF were 14.0 ± 3.85 and 8.0 8 ± 1.2 7 (P <0 .0 1). Conclusion: 1 92 Ir implantation is an effective method for the treatment of oral squamous cell carcinoma. The detection of DNA content in oral squamous cell carcinoma can be used to measure the effect of radiation therapy and help to make the treatment plan.
