目的观察肝细胞生长因子(HGF)联合吡柔比星(Theprubicin,THP)体外对肝癌细胞(HepG2)化疗是否有协同作用。方法肝癌细胞处理分为单用组:THP(20μg/L)和HGF(20μg/L);联合组:THP(20μg/L)+HGF(20μg/L);生理盐水对照组。MTT法检测细胞活性,流式细胞仪测定细胞凋亡。结果所有试验组肝癌细胞生长抑制率及凋亡率均显著高于对照组。HGF和THP协同处理后,癌细胞生长抑制率及凋亡与坏死比例[(53.86±5.53)%,(33.06±6.62)%]与单用组HGF[(31.27±3.86)%,(18.16±1.71)%]或THP[(32.11±4.19)%,(13.16±3.74)%]相比显著升高(P<0.05)。结论THP联合HGF体外作用于肝癌细胞,能够抑制其生长并诱导其凋亡,且具有协同作用。
Objective To observe whether hepatocyte growth factor (HGF) combined with Theprubicin (THP) has a synergistic effect on HepG2 chemotherapy in vitro. Methods Liver cancer cells were divided into three groups: THP (20μg / L) and HGF (20μg / L); the combination group: THP (20μg / L) and HGF (20μg / L); saline control group. Cell viability was measured by MTT assay and apoptosis by flow cytometry. Results The growth inhibition rate and apoptosis rate of hepatoma cells in all experimental groups were significantly higher than those in control group. After treatment with HGF and THP, the rate of tumor growth inhibition and the ratio of apoptosis and necrosis [(53.86 ± 5.53)% vs (33.06 ± 6.62)%] and HGF (31.27 ± 3.86)% and (18.16 ± 1.71) )%] Or THP [(32.11 ± 4.19)%, (13.16 ± 3.74)%] (P <0.05). Conclusion THP combined with HGF can inhibit the growth of hepatocellular carcinoma cells in vitro and induce its apoptosis with synergistic effect.
