目的探讨左金丸药物血清对人乳腺癌MCF-7细胞周期和凋亡的影响。方法制备左金丸药物血清,以MTT比色法检测5%、10%和20%左金丸药物血清在24h、48h和72h对MCF-7细胞的增殖抑制作用;流式细胞仪检测细胞周期和凋亡;Western blot法检测Cyclin D1、Cyciln E、CDK2、CDK4、p21、p27、p53和Bax的表达。结果与无药物血清比较,左金丸药物血清显著抑制MCF-7细胞的增殖(P<0.05),以10%浓度48h给药效果最好。与无药物血清组(Sub-G_1期细胞:2.13%±1.83%;G_0/G_1期细胞:39.04%±6.84%)比较,10%左金丸药物血清组(Sub-G_1期细胞:12.47%±1.82%;G_0/G_1期细胞:46.80%±3.00%),显著增加了Sub-G1期和G_0/G_1期的细胞数(P<0.01)。而且,左金丸药物血清下调了Cyclin D1,Cyciln E表达,并上调了p21,p27,p53和Bax表达。结论左金丸药物血清具有抑制MCF-7人乳腺癌细胞增殖作用,其机制可能与阻滞细胞G0/G1周期并诱导细胞凋亡有关。
Objective To investigate the effect of Zuojin Pill serum on the cell cycle and apoptosis of human breast cancer cell line MCF-7. Methods Zuojinwan serum was prepared by MTT colorimetric assay. The inhibitory effects of 5%, 10% and 20% Zuojinwan serum on the proliferation of MCF-7 cells were detected by MTT assay. The cell cycle and apoptosis were detected by flow cytometry The expressions of Cyclin D1, Cyciln E, CDK2, CDK4, p21, p27, p53 and Bax were detected by Western blot. Results Compared with no drug serum, Zuojin Pill serum significantly inhibited the proliferation of MCF-7 cells (P <0.05), and the best effect was obtained at 48% of 10% concentration. Compared with the drug-free serum group (Sub-G_1 phase cells: 2.13% ± 1.83%; G_0 / G_1 phase cells: 39.04% ± 6.84%), 10% %; G_0 / G_1 cells: 46.80% ± 3.00%) significantly increased the number of cells in the sub-G1 and G_0 / G_1 phases (P <0.01). Moreover, Zuojin Pill serum lowered Cyclin D1, Cyciln E expression and up-regulated p21, p27, p53 and Bax expression. Conclusion Zuojinwan serum can inhibit the proliferation of MCF-7 human breast cancer cells. The mechanism may be related to arresting G0 / G1 cycle and inducing apoptosis.
