目的通过测定2002~2003年河北省鼠疫自然疫源地分离的27株鼠疫菌,得到其生化特性的本底资料。方法采用液体汤管法和固体试管法(斜面或高层)实验测定27株鼠疫菌对糖醇的酵解能力和生化反应。结果(1)27株鼠疫菌对甘油、L阿胶糖等多种糖酵解,产酸不产气;不发酵麦芽糖、鼠李糖,72h内对密二糖不酵解,5d以后有8株菌的部分糖管呈迟缓发酵。(2)生化反应:27株鼠疫菌在还原KNO3、硝化反应、VP试验、尿素试验、靛基质试验呈阴性,不液化明胶,在无蛋白胨酸性琼脂培基上不生长,不能利用枸橼酸盐;甲基红反应阳性,均能轻微产生H2S。结论被试的27株鼠疫菌按中国鼠疫菌生态型分型标准属于B群鄂尔多斯高原型。主要生化特征稳定,与疫源地过去分离的鼠疫菌株生化特性基本一致。
Objective To determine the biochemical characteristics of 27 strains of Yersinia pestis isolated from the natural foci of plague in Hebei province from 2002 to 2003. Methods The ability of Yersinia pestis to ferment sugar and the biochemical reaction of 27 strains of Yersinia pestis were determined by liquid tube method and solid tube method (inclined plane or high layer). Results (1) 27 strains of Yersinia pestis did not produce maltose and rhamnose on a variety of glycolysis such as glycerol and L-guar gum, Part of the bacteria was slow fermentation of sugar tube. (2) Biochemical reaction: 27 strains of Yersinia pestis reduced KNO3, nitrification reaction, VP test, urea test, indomethacin test negative, non-liquefied gelatin, no peptone acid agar medium does not grow, can not use citrate ; Methyl red reaction was positive, can produce mild H2S. Conclusion The 27 strains of Yersinia pestis belong to Group B Erdos Plateau according to the Chinese Yersinia pestis ecotype classification criteria. The main biochemical characteristics are stable and the biochemical characteristics of the plague strains isolated from the past in the past are basically the same.
