目的探讨益智方对阿尔茨海默病(AD)大鼠学习记忆的作用及海马神经细胞凋亡相关基因表达的影响。方法在基础饲料中添加三氯化铝,Al3+剂量为111.9 mg/kg体重,连续染毒3个月复制大鼠AD模型,同时给大鼠灌服不同剂量的益智方中药,采用Y-电迷宫仪检测大鼠学习记忆能力,流式细胞检测大鼠海马神经细胞Bcl-2、Bax、p53表达。结果益智方能明显增加AD大鼠逃避正确率、缩短逃避潜伏期,上调(促进)AD大鼠海马神经细胞Bcl-2表达,下调(抑制)Bax、p53表达。结论益智方能明显改善AD大鼠的学习记忆能力,其机制可能与益智方能上调AD大鼠海马神经细胞Bcl-2表达,下调Bax、p53表达有关。
Objective To investigate the effect of Yizhi Recipe on learning and memory of Alzheimer’s disease (AD) rats and the expression of apoptosis related genes in hippocampal neurons. Methods AlCl3 was added into the basal diet, the dose of Al3 + was 111.9 mg / kg body weight, and the model of AD was induced by continuous exposure for 3 months. At the same time, Labyrinth apparatus was used to detect the learning and memory abilities of rats. Flow cytometry was used to detect the expression of Bcl-2, Bax and p53 in rat hippocampal neurons. Results Yizhi Decoction could significantly increase the escape rate, shorten the escape latency, up-regulate the expression of Bcl-2 and down-regulate the expression of Bax and p53 in hippocampal neurons of AD rats. Conclusion Yizhi Decoction can significantly improve the learning and memory abilities of AD rats, which may be related to the fact that Yizhi Decoction can up-regulate the expression of Bcl-2 and down-regulate the expression of Bax and p53 in hippocampal neurons of AD rats.
