为防止重瓣矮牵牛(Petunia hybrida Vilm)试管苗倒伏,提高其移栽成活率,研究了生根培养时使用B_9和PP_(333)的效果。发现采用B_9和PP_(333) 30ppm在生根培养后4天,对新梢进行喷洒处理,可以改善发根情况,处理苗移栽成活率可大大提高,且叶色深绿。组织学观察表明,重瓣矮牵牛新梢根的发生有二种方式。B_9处理对根的分化无影响,PP_(333)处理不影响新梢切口端脱分化形成分生细胞团,但抑制了分生细胞团再分化根。B_9处理不影响过氧化物酶同功酶谱带数,但稍增加了过氧化物酶活性,PP_(333)处理增加了一条同功酶谱带,且显著地增加了过氧化物酶活性。
In order to prevent the lodging of plantlets of Petunia hybrida Vilm and improve the transplanting survival rate, the effect of using B_9 and PP_ (333) in rooting culture was studied. It was found that spray treatment of shoots with B_9 and PP_ (333) 30 ppm for 4 days after rooting could improve the condition of hair root. The survival rate of the treated seedlings could be greatly improved and the leaf color was dark green. Histological observation showed that there are two ways to produce root tip of Petunia hybrida. B_9 treatment had no effect on root differentiation. PP_ (333) treatment did not affect the dedifferentiation of shoots at the shoot tip to form meristematic cells, but inhibited the redifferentiation of meristematic cells. B_9 treatment did not affect the peroxidase isozyme bands, but slightly increased the peroxidase activity, PP_ (333) treatment increased an isozyme band, and significantly increased peroxidase activity.
