目的研究米非司酮对人早孕绒毛组织中转化生长因子β(TGF-β)信号传导通路功能状态的影响以及与终止妊娠之间的关系。方法用免疫组化SP法和RT-PCR方法分别检测药物流产组(20例)和人工流产组(13例)绒毛组织中TGF-β信号传导通路的下游靶基因smad7的蛋白质和mRNA表达水平,结果用平均光密度值和光度比值表示,半定量分析结果。结果药物流产组和人工流产组绒毛组织中smad7的免疫组化结果的平均光密度值分别为0.346 6±0.046 9和0.233 7±0.065 7,P<0.01。药物流产组和人工流产组绒毛组织中smad7mRNA的平均光度比值为0.561 7±0.410 3和0.222 2±0.244 1,P<0.02。结论米非司酮能够增强TGF-β信号传导通路的功能,使该信号通路的下游靶基因纤溶酶原激活物抑制物表达增多,降低了滋养细胞的转移、侵袭能力,可能是该药物致早孕流产的机制之一。
Objective To investigate the effect of mifepristone on the functional status of TGF-β signal transduction pathway in human chorionic villi in early pregnancy and its relationship with termination of pregnancy. Methods Immunohistochemical SP method and RT-PCR method were used to detect the protein and mRNA expression levels of smad7, a downstream target of TGF-β signaling pathway, in villus tissues of 20 cases of abortion group and 13 cases of induced abortion group. The results with the average optical density value and photometric ratio, semi-quantitative analysis of the results. Results The average optical density of Smad7 in the villus tissue of abortion group and abortion group were 0.346 6 ± 0.046 9 and 0.233 7 ± 0.065 7, respectively (P <0.01). The mean luminosity ratios of smad7mRNA in villus tissues of abortion group and abortion group were 0.561 7 ± 0.410 3 and 0.222 2 ± 0.244 1, P <0.02. Conclusion Mifepristone can enhance the function of TGF-β signal transduction pathway, increase the expression of plasminogen activator inhibitor downstream target gene of the signal pathway, reduce the ability of trophoblast metastasis and invasion, which may be caused by the drug induced One of the mechanisms of early pregnancy miscarriage.
