目的 研究DNA聚合酶 β基因在采自食管癌高发区的食管癌标本中的突变情况。 方法 提取总RNA反转录合成cDNA ,经PCR扩增后以SSCP检测其变异情况。结果 在食管癌组织标本中存在polβ基因的突变 :2 0例癌组织标本中发现 9例突变 (45 %、9/ 2 0 ) ,癌旁组织中发现 1例突变 (5 %、1 / 2 0 )。结论 在食管癌组织标本中确实存在polβ基因的突变 ,推测在食管癌的的发生发展过程中polβ基因突变是一个始动因素 ,是各种致癌因子作用的枢纽 ,由于该基因的突变可能使得癌基因、抑癌基因如 :ras、p53、Rb等的原发性损伤得不到及时正确修复 ,导致细胞周期的失控、增殖过多 ,促进恶性转化
Objective To study the mutation of DNA polymerase β gene in esophageal cancer samples collected from high incidence area of esophageal cancer. Methods Total RNA was extracted and reverse transcribed into cDNA. The variation of the cDNA was detected by SSCP after PCR amplification. Results There was a mutation of polβ gene in esophageal cancer specimens: 9 mutations (45%, 9/20) were found in 20 specimens of cancerous tissue and 1 mutation (5%, 1/2 0) in adjacent tissues ). Conclusion The mutation of polβ gene does exist in esophageal cancer tissue samples. It is speculated that the mutation of polβ gene is an initiating factor in the development and progression of esophageal cancer, which is the pivotal role of various oncogenic factors. Because mutations of this gene may cause cancer Primary damage to genes, tumor suppressor genes such as ras, p53, and Rb is not repaired in time, resulting in loss of control of the cell cycle, hyperproliferation, and promotion of malignant transformation
