肝片吸虫是一种引起哺乳动物片吸虫病的肝内吸虫,寄生于胆管,主要进行无氧代谢,通常认为宿主可借助氧自由基杀伤虫体。超氧化物岐化酶(SOD)是一种重要的抗氧化酶,在肝片吸虫抵抗宿主氧自由基介导的杀伤机制中对虫体起保护作用。为进一步阐明其机制,作者从感染肝片吸虫的牛肝内收集成虫,抽提SOD,经纯化后分析其特性,并建立cDNA文库;克隆其基因,在大肠杆菌中表达。
Fasciola hepatica is a type of liver fluke that causes mammalian chelonia, parasites in the bile duct, mainly for anaerobic metabolism, and is often thought to kill the worms with oxygen free radicals. Superoxide dismutase (SOD) is an important antioxidant enzyme that plays a protective role against parasites in the presence of oxygen free radical-mediated killing of Fasciola hepatica. To further elucidate its mechanism, the authors collected adult worms from bovine liver infected with Fasciola hepatica and extracted SOD. After purification, their characteristics were analyzed and a cDNA library was constructed. The gene was cloned and expressed in E. coli.
