目的:测量心肌缺血再灌注损伤(myocardial ischemic reperfusion injury,MIRI)后大鼠血清中的血脂、心肌酶和心肌匀浆中的丙二醛(malondialdehyde,MDA)和相关抗氧化酶的含量。观察调脂通络胶囊可能的心肌保护作用和相关的机制。方法:本实验在高脂血症模型的基础上建立MIRI模型,将56只大鼠分为调脂通络胶囊高、中、低剂量组(8.64,4.32,2.16 g·kg-1·d-1)、高脂模型组、非高脂模型组、阿托伐他汀阳性对照组和高脂假手术组,每组8只大鼠。喂食4周高脂饲料(非高脂模型组正常饮食)后药物干预或生理盐水灌胃4周,结扎左冠状动脉前降支30 min(假手术组只穿线不结扎),松线再灌注24 h后采血测定血清血脂(TC,TG,HDL,LDL)含量,血清心肌酶(LDH,CK-MB)活性;取缺血区心肌组织测定大鼠心肌匀浆中抗氧化酶[超氧化物歧化酶(SOD),谷胱甘肽过氧化物酶(GSH-Px),过氧化氢酶(CAT)]活性,MDA的含量。结果:与正常饮食组相比,高脂模型组的血脂、心肌酶、MDA明显升高,抗氧化酶含量明显降低;各药物组均可以明显降低心肌酶和MDA含量、升高抗氧化酶SOD,CAT,GSH-Px的活性,调脂通络胶囊各剂量组与他汀组无统计学差异。结论:调脂通络胶囊和阿托伐他汀具有减少自由基聚集,提高抗氧化酶活力的作用。
OBJECTIVE: To measure the levels of malondialdehyde (MDA) and related antioxidant enzymes in serum lipids, myocardial enzymes and myocardial homogenates of myocardial ischemic reperfusion injury (MIRI) in rats. To observe the possible myocardial protective effect of Tiaozhi Tongluo Capsule and its related mechanism. Methods: MIRI model was established on the basis of hyperlipidemia model. 56 rats were divided into high, medium and low doses of Tiaozhi Tongluo Capsules (8.64,4.32,2.16 g · kg-1 · d- 1), hyperlipidemic model group, non-hyperlipidemic model group, atorvastatin positive control group and hyperlipidemic sham operation group, 8 rats in each group. After 4 weeks of high-fat diet (normal diet of non-hyperlipidemic model group), drug intervention or saline was given for 4 weeks. Ligation of the left anterior descending coronary artery for 30 minutes (sham-operated group only without threading) (TC, TG, HDL, LDL) and serum LDH (CK-MB) were measured by ELISA. The activities of antioxidant enzymes [superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT)] activity and MDA content. Results: Compared with the normal diet group, the levels of lipids, myocardial enzymes and MDA were significantly increased and the contents of antioxidant enzymes were significantly decreased in the high-fat diet group. The levels of myocardial enzymes and MDA, and the levels of antioxidant enzymes SOD , CAT, GSH-Px activity, Tiaolijutongluo capsule dose group and statin group was not statistically different. Conclusion: Tiaozhi Tongluo Capsule and atorvastatin have the effects of reducing free radical accumulation and increasing antioxidant enzyme activity.
