A previously unknown acute leukemia subtype with both myeloid and natural killer cell characteristics was tested and described herein. The authors measured 20 (accounting for 6%) of 350 adult AML patients. They all had consistent immunophenotypes of CD33+, CD56+, CD11a+, CD13 (low expression), CD15 (low expression), and CD34. ~±, HLA-DR~-, CD16~-. Using multicolor flow cytometric analysis, the results demonstrated more strongly the simultaneous expression of myeloid differentiation antigens (CD33, CD13, CD15) and NK cell-associated antigens (CD56). Reverse transcription PCR (Rt-PCR) analysis also fully demonstrated the consistency of CD56 in leukemic naive cells. Except for 2 cases of CD4 expression, none of them expressed CD2, CD3, or CD8, and no TCR gene rearrangement was detected.
