One of the four methods used to determine the accuracy of porcine pancreatic elastase at low concentrations was one synthetic substrate and the other three were native substrate elastin. The most accurate method uses the synthetic substrate Succ (Ala) _3NA (intra-assay coefficient of variation = 1.9%, inter-assay variation Coefficient = 2.5%); the most accurate method using elastin as a substrate was fluorescein-labeled elastin (intra-assay CV = 5.1%, inter-assay CV = 5.5%).
