目的:观察镰形棘豆对人肝癌细胞SMMC-7721的增殖及其基质金属蛋白酶-2(MMP-2)分泌量的影响。方法:以不同质量浓度(25,50,100,250,500 mg.L-1)的镰形棘豆挥发油、总多糖、总黄酮、总生物碱和总皂苷部位处理SMMC-7721细胞24 h后,采用MTT法检测各部位对SMMC-7721细胞增殖的抑制作用;采用ELISA法测定挥发油部位和总黄酮部位处理SMMC-7721肝癌细胞24 h后细胞上清中MMP-2的分泌量;Real-time PCR法检测处理24 h后MMP-2 mRNA的转录情况。结果:MTT实验结果表明,挥发油和总黄酮部位抑制SMMC-7721细胞的增殖,并表现出一定的浓度依赖性。ELISA实验表明挥发油部位显著性地抑制MMP-2分泌量;Real-time PCR实验表明挥发油和总黄酮均显著抑制MMP-2 mRNA的表达。结论:镰形棘豆挥发油和总黄酮部位抑制人肝癌细胞SMMC-7721的增殖活性,可能与其下调MMP-2的分泌量和转录水平有关。
Objective: To observe the effects of Oxytropis falcate on the proliferation and the secretion of matrix metalloproteinase-2 (MMP-2) in human hepatocellular carcinoma cell line SMMC-7721. Methods: SMMC-7721 cells were treated with volatile oil, total polysaccharides, total flavonoids, total alkaloids and total saponins of different concentrations (25,50,100,250,500 mg.L-1) for 24 h. MTT assay The inhibition of SMMC-7721 cell proliferation was measured by ELISA. The secretion of MMP-2 in supernatant of SMMC-7721 hepatoma cells treated with volatile oil and total flavonoids was determined by ELISA. The expression of MMP-2 in supernatant of SMMC- Post-MMP-2 mRNA transcription. Results: MTT results showed that the volatile oil and flavonoids inhibited the proliferation of SMMC-7721 cells in a concentration-dependent manner. ELISA experiments showed that the volatile oil fraction significantly inhibited the secretion of MMP-2; Real-time PCR experiments showed that volatile oil and total flavonoids significantly inhibited the expression of MMP-2 mRNA. CONCLUSION: The inhibitory effects of volatile oil and total flavonoids of Oxya Siniad on the proliferation of human hepatocellular carcinoma cell line SMMC-7721 may be related to the downregulation of MMP-2 secretion and transcription.
