目的探究上调SK-Hep1肝癌细胞中lnc RNA-AK058003的表达对肝癌裸鼠移植瘤生长的影响。方法采用慢病毒转染的方法将lnc RNA-AK058003的重组质粒和空质粒分别转入SK-Hep1肝癌细胞株中,经新霉素筛选出lnc RNAAK058003稳定过表达的细胞株,用定量PCR的方法检测lnc RNA-AK058003在SK-Hep1肝癌细胞中的过表达,并且将稳定过表达lnc RNA-AK058003的SK-Hep1肝癌细胞和含有空质粒的SK-Hep1肝癌细胞分别注射入裸鼠腋下皮下,定期测量裸鼠移植瘤的长径和短径,从而计算出移植瘤的体积,饲养35天后测量移植瘤的重量。结果构建稳定过表达lnc RNA-AK058003的肝癌细胞株,裸鼠皮下荷瘤实验结果显示,过表达组裸鼠移植瘤的体积(308.4±439.4mm~3)、重量(0.464±0.518g)和生长速度均低于空质粒组(1410.0±973.0mm~3,1.363±0.856g,P<0.05)。结论 lnc RNA-AK058003对肝癌裸鼠移植瘤的增殖具有抑制作用。
Objective To investigate the effect of up-regulating the expression of lnc RNA-AK058003 in SK-Hep1 hepatoma cells on the growth of hepatocellular carcinoma xenografts in nude mice. Methods Recombinant plasmid and empty plasmid of lnc RNA-AK058003 were transfected into SK-Hep1 hepatocellular carcinoma cell line by lentiviral transfection. The cell lines stably overexpressing lncRNAAK058003 were screened by neomycin and quantified by quantitative PCR The overexpression of lnc RNA-AK058003 in SK-Hep1 hepatoma cells was detected, and SK-Hep1 hepatoma cells stably overexpressing lnc RNA-AK058003 and SK-Hep1 hepatoma cells containing empty plasmids were injected subcutaneously into nude mice respectively. The long and short diameters of nude mice xenografts were measured regularly to calculate the volume of the xenografts and the weight of the xenografts was measured after 35 days of feeding. Results The constructed hepatoma cell line stably overexpressing lnc RNA-AK058003 showed that the tumor volume in nude mice (308.4 ± 439.4mm ~ 3), weight (0.464 ± 0.518g) and growth The speed was lower than the empty plasmid group (1410.0 ± 973.0mm ~ 3,1.363 ± 0.856g, P <0.05). Conclusion lnc RNA-AK058003 can inhibit the proliferation of hepatocellular carcinoma xenografts in nude mice.
