目的:研究川芎提取物对硝酸甘油所致偏头痛大鼠行为学症状及血浆、脑组织中的5-羟色胺(5-HT)、多巴胺(DA)、去甲肾上腺素(NE)含量的影响,初步筛选川芎治疗偏头痛的有效部位。方法:将川芎乙醇总提取物,采用系统溶剂法依次以氯仿、乙酸乙酯、正丁醇萃取,得到各提取部位后,分别灌胃给予实验大鼠。采用硝酸甘油致大鼠偏头痛模型,观察给药后大鼠耳红、挠头和爬笼次数等行为学指标;高效液相色谱-荧光检测法测定各组大鼠血浆及脑组织中的5-HT、DA、NE含量。结果:川芎乙醇提取物、乙酸乙酯提取物能显著抑制偏头痛大鼠耳红消失时间和挠头、爬笼次数;并能不同程度升高偏头痛大鼠血及脑内5-HT、DA、NE含量(P<0.05、0.01),且两者间无显著性差异。结论:乙酸乙酯提取部位是川芎治疗偏头痛的有效部位。
Objective: To investigate the effects of Chuanxiong Extract on behavioral symptoms and the content of 5-HT, DA and NE in plasma and brain of migraine rats induced by nitroglycerin, Chuanxiong initial screening of effective parts of migraine. Methods: The total ethanol extract of Chuanxiong rhizome was extracted with chloroform, ethyl acetate and n-butanol by systematic solvent method. After the extraction sites were obtained, the experimental rats were administered intragastrically. The model of migraine induced by nitroglycerin was used to observe the behavioral indexes such as the number of ear red, scratching head and climbing cage in rats. The contents of 5- HT, DA, NE content. Results: Chuanxiong ethanol extract, ethyl acetate extract can significantly inhibit migraine rat ear red disappearance of time and scratching, climbing cage number; and to varying degrees, increased blood and brain migraine rat 5-HT, DA, NE content (P <0.05,0.01), and no significant difference between the two. Conclusion: Ethyl acetate extraction site is an effective part of Chuanxiong for the treatment of migraine.
