目的:优选麝香总多肽的研磨提取工艺,为麝香的品种鉴定与质量评价提供参考。方法:采用福林酚法测定麝香总多肽含量。以麝香总多肽含量为指标,通过正交试验考察提取时间、溶媒种类及用量对提取工艺的影响。结果:最佳提取工艺为三羟甲基氨基甲烷-盐酸缓冲液(p H 8.0,0.025 mol·L-1),料液比1∶100,研磨时间3 h,放置过夜;麝香总多肽平均质量分数达20.71%,经冷冻干燥技术处理后麝香总多肽收率62.4%。结论:优选的提取工艺稳定可行,获得麝香总多肽的提取率较高,为麝香的品质分析提供实验依据。
OBJECTIVE: To optimize the grinding and extraction process of total musk peptide to provide references for the identification and quality evaluation of Musk. Methods: Determination of total flavonoids in musk by Folin phenol method. Taking the total musk content of musk as an index, the effects of extraction time, solvent type and dosage on the extraction process were investigated by orthogonal test. Results: The best extraction process was Tris-HCl buffer (p H 8.0, 0.025 mol·L-1), the ratio of solid to liquid was 1: 100, the milling time was 3 h and left overnight. The average mass of total musk The score reached 20.71%. The total musk yield was 62.4% after freeze-drying. Conclusion: The optimal extraction process is stable and feasible, and the extraction rate of total musk is higher, which provides the experimental basis for the quality analysis of musk.
