目的:检测miR-136对胃癌细胞SGC7901增殖和凋亡的影响及其机制。方法:分别利用MTT实验和Annex in V-PI实验检测转染miR-136 inhibitor的胃癌细胞SGC7901增殖和凋亡。利用western blot和双荧光素酶报告基因技术检测miR-136的靶基因。结果:抑制miR-136表达可以明显抑制SGC7901细胞增殖和诱导其凋亡。miR-136能够与PPP2R2A mRNA 3’-UTR区结合。结论:抑制miR-136表达能够促进PPP2R2A蛋白表达,进而引起胃癌细胞凋亡。
Objective: To investigate the effect and mechanism of miR-136 on proliferation and apoptosis of gastric cancer cell line SGC7901. Methods: The proliferation and apoptosis of gastric cancer cell line SGC7901 transfected with miR-136 inhibitor were detected by MTT assay and Annexin V-PI assay respectively. The target gene of miR-136 was detected by western blot and dual-luciferase reporter assay. Results: Inhibition of miR-136 expression significantly inhibited SGC7901 cell proliferation and induced its apoptosis. miR-136 is able to bind to the PPP2R2A mRNA 3’-UTR region. Conclusion: Inhibition of miR-136 expression can promote the expression of PPP2R2A protein and induce gastric cancer cell apoptosis.
