结直肠癌是威胁人类健康的常见肿瘤之一,目前缺乏敏感度、特异度高的早期诊断方法。蛋白质组学为结直肠癌许多研究领域拓开了新视野,表面增强激光解吸离子化飞行时间质谱(SELDI-TOF-MS)是蛋白质芯片和质谱技术的结合,主要由蛋白质芯片、芯片阅读器和生物信息学3大部分组成,可直接分析患者血清、淋巴液、脑脊液、尿液、细胞分泌液等样品而无需复杂的样品制备,具有快速、简便、易行高通量平行分析等特点,是蛋白质组学的主要技术平台之一,目前已经初步应用于结直肠癌早期诊断、术后随访及标志物鉴定等方面的研究。作者综述了SELDI-TOF-MS的基本原理、技术特点及其在结直肠癌蛋白质组学研究中的应用和展望。

Surface-enhanced laser desorption ionization time-of-flight mass spectrometry (SELDI-TOF-MS) is a combination of protein chip and mass spectrometry technology that consists mainly of protein chips, chip readers and Bioinformatics 3 most of the components can be directly analyzed in patients with serum, lymph, cerebrospinal fluid, urine, cell exudates and other samples without the need for complex sample preparation, with fast, easy, easy parallel high-throughput analysis features Proteomics, one of the major technology platforms, has now been initially applied in early diagnosis of colorectal cancer, postoperative follow-up and identification of markers and other aspects of research. The authors summarize the basic principles and technical characteristics of SELDI-TOF-MS and its application and prospect in proteomics of colorectal cancer.
