目的考察复方蒲黄提取物对肾炎性模型大鼠血尿及肾功能的影响,对复方蒲黄提取物进行药效学评价,为开发医院制剂复方蒲黄颗粒提供实验基础。方法采用免疫原牛血清白蛋白(BSA)灌胃、四氯化碳(CCl4)皮下注射、脂多糖(LPS)尾静脉注射建立肾炎性血尿大鼠模型后,高、中、低剂量复方蒲黄提取物灌胃给药6周,分别于第4周、第6周测定大鼠血肌酐(SCr)和尿素氮(BUN)浓度、血清总蛋白(TP)、24 h尿蛋白、尿肌酐定量、尿畸形红细胞计数,计算内生肌酐清除率(CCr)。结果蒲黄组第4、6周分别与模型对照组比较,能明显减少尿中畸形红细胞个数、24 h尿蛋白、尿肌酐定量,降低血肌酐和尿素氮值,提高血清总蛋白以及内生肌酐清除率,差异有显著性(P<0.01或P<0.05)。结论复方蒲黄提取物可以减少肾炎性血尿模型大鼠的血尿、蛋白尿症状,改善肾功能各项指标,以高剂量组作用效果最佳。
Objective To investigate the effect of compound Puhuang extract on hematuria and renal function of rats with nephritis model, evaluate the pharmacodynamics of compound Puhuang extract, and provide experimental basis for the development of Puhuang granules of hospital preparation. Methods The rat model of nephritis hematuria was established by intraperitoneal injection of BSA, subcutaneous injection of CCl4 and tail vein injection of lipopolysaccharide (LPS). High, medium and low doses of Compound Puhuang The rats were administered intragastrically for 6 weeks. The levels of serum creatinine (SCr) and blood urea nitrogen (BUN), serum total protein (TP), 24 h urine protein and urinary creatinine were measured at the 4th and 6th week respectively. Urinary malformations, red blood cell count, calculate endogenous creatinine clearance (CCr). Results Compared with the model control group, the numbers of abnormal red blood cells, 24-hour urinary protein, urinary creatinine, urinary creatinine, serum creatinine, serum creatinine and urea nitrogen were significantly decreased at 4 and 6 weeks in Puhuang group, Creatinine clearance rate, the difference was significant (P <0.01 or P <0.05). Conclusion Compound Puhuang extract can reduce the hematuria, proteinuria symptoms of renal nephritis hematuria model rats, improve various indicators of renal function in high-dose group best effect.
