目的研究慢性酒精中毒大鼠神经细胞凋亡及Fas、Caspase8蛋白表达情况。方法逐步增加饮水中酒精浓度,建立慢性酒精中毒大鼠动物模型,HE染色光镜下观察大鼠脑组织病理变化,Tunnel法检测大鼠脑细胞凋亡情况,免疫组化法检测Fas、Caspase8蛋白表达情况。结果酒精组大脑皮质、海马、小脑等部位均有神经细胞数目缺失及细胞变性和损伤,酒精组各脑区凋亡细胞数和Fas、Casepse8蛋白表达阳性细胞数均高于对照组,两组比较差异有统计学意义(P<0.05)。结论慢性酒精中毒大鼠脑存在明显神经细胞凋亡,伴有Fas、Caspase-8蛋白表达增加,提示细胞死亡受体途径在慢性酒精中毒大鼠脑损伤病理过程中发挥作用。
Objective To investigate the apoptosis of neurons and expression of Fas and Caspase8 in rats with chronic alcoholism. Methods The alcohol concentration in drinking water was gradually increased to establish animal models of chronic alcoholism. The pathological changes of brain tissue were observed under light microscope with HE staining. The apoptosis of brain cells was detected by Tunnel method. The expressions of Fas and Caspase8 proteins were detected by immunohistochemistry. Express the situation. Results The number of neurons and the degeneration and injury of cells in the cerebral cortex, hippocampus and cerebellum of alcohol group were decreased. The number of apoptotic cells and the expression of Fas and Casepse8 in alcohol group were higher than those in control group The difference was statistically significant (P <0.05). Conclusion There is obvious neuronal apoptosis in the brain of rats with chronic alcoholism, accompanied with an increase of Fas and Caspase-8 protein expression, suggesting that the cell death receptor pathway plays a role in the pathological process of brain injury in chronic alcoholism rats.
