目的 研究肠三叶因子 (ITF)及其mRNA在大鼠肠道的分布规律。方法 采用原位杂交、免疫组化等手段观察了ITF及ITFmRNA在大鼠肠道的表达并使用高效液相色谱仪检测了不同肠段ITF的含量。结果 从十二指肠至结肠均有ITF及ITFmRNA表达 ,主要分布于肠绒毛杯状细胞 ,其中ITFmRNA仅在杯状细胞的基底和边缘表达 ,其它区域的杯状细胞则呈ITFmRNA阴性反应 ,同时发现部分其它部位的肠上皮细胞中亦有少量ITF及ITFmRNA表达。ITF定量分析显示 ,整个肠道中以十二指肠含量最高 ,空回肠次之 ,结肠含量较低 ,各段小肠中ITF含量差异不显著 (P >0 .0 5 ) ,但都明显高于结肠 (P <0 .0 1)。结论 杯状细胞是合成ITF的主要场所 ,ITF在肠道分布的差异可能同各肠段分泌的粘液性质不同有关。
Objective To study the distribution of intestinal trefoil factor (ITF) and its mRNA in rat intestine. Methods The expressions of ITF and ITF mRNA in the intestine of rats were observed by in situ hybridization and immunohistochemistry, and the content of ITF in different intestinal segments was detected by high performance liquid chromatography. Results Both ITF and ITF mRNA were expressed in the duodenum and colon, which were mainly distributed in goblet goblet cells, of which ITF mRNA was only expressed on the basal and marginal goblet cells, and goblet cells in other areas showed ITF mRNA negative reaction A small amount of ITF and ITF mRNA expression was also found in some other parts of intestinal epithelial cells. ITF quantitative analysis showed that the duodenum was the highest in the whole intestine, the second was in the ileum, the lower in the colon, and the ITF content in the small intestine was not significantly different (P> 0.05), but both were significantly higher than those in the colon (P <0. 01). Conclusion Goblet cells are the main sites for the synthesis of ITF. The differences in the distribution of ITF in the intestine may be related to the different mucus secretions in each intestine.
