目的:观察补中益气汤含药血清对转化生长因子-β1(transforming growth factor-β1,TGF-β1)介导的人肺腺癌A549细胞上皮间质转化过程中Twistl表达的影响。方法:制备最佳浓度补中益气汤最佳浓度含药血清作用于TGF-β1诱导的A549细胞,实验分组为空白血清组、空白血清+TGF-β1组、以及补中益气汤最佳浓度补中益气汤含药血清+TGF-β1组。采用免疫组化、western-blot和realtime PCR法分别检测Twistl在蛋白和基因水平表达的变化。结果:最佳浓度的补中益气汤含药血清能使上皮间质化的A549细胞中Twistl蛋白表达下降,mRNA表达水平降低。结论:补中益气汤含药血清能抑制TGF-β1介导的A549细胞上皮间质化过程中Twistl表达。
OBJECTIVE: To observe the effect of Buzhong Yiqi decoction-containing serum on the expression of Twistl in human lung adenocarcinoma A549 cells induced by transforming growth factor-β1 (TGF-β1). Methods: The optimal concentrations of Bu Zhong Yi Qi decoction-containing serum were given to A549 cells induced by TGF-β1. The experimental groups were blank serum group, blank serum + TGF-β1 group and Buzhong Yiqi Decoction Concentration Bu Zhong Yi Qi Tang containing serum + TGF-β1 group. Immunohistochemistry, western-blot and realtime PCR were used to detect the expression of Twistl at the protein and gene level respectively. Results: The optimal concentrations of Buzhong Yiqi decoction serum could decrease the expression of Twistl protein and decrease the mRNA expression level of epithelial mesenchyme in A549 cells. Conclusion: Buzhong Yiqi decoction containing serum can inhibit Twistl expression in TGF-β1-mediated epithelial-mesenchymal transition of A549 cells.
