目的通过观察乌司他丁对多器官功能障碍综合征(MODS)大鼠血清、肾组织中细胞因子的影响及肾脏病理改变,探讨其对肾脏的保护作用。方法将36只大鼠随机分为正常对照组、内毒素(LPS)组、LPS+乌司他丁组。舌下静脉注射LPS方法制作MODS模型,治疗组在给予LPS前10min,通过舌下静脉注射乌司他丁,分别于给药后2h、6h每组各杀死6只,同时取肾组织制备匀浆并作病理学检查。采用放射免疫法测定血清及肾组织中TNFα、IL6、IL1β、iNOS。结果LPS+乌司他丁组血清及肾组织TNFα、IL6、IL1β、iNOS较LPS组明显降低(P<0.05),病理改变明显轻于LPS组。结论乌司他丁能显著减少MODS大鼠血清及肾组织中TNFα、IL6、IL1β、iNOS的含量,减轻炎性细胞因子介导的肾脏损伤,保护肾脏功能。
Objective To observe the effects of ulinastatin on the serum and renal cytokines in rats with multiple organ dysfunction syndrome (MODS) and the renal pathological changes, and to explore its protective effect on the kidney. Methods Thirty-six rats were randomly divided into normal control group, endotoxin (LPS) group and LPS + ulinastatin group. Sublingual vein injection of LPS method for the production of MODS model, the treatment group 10 minutes before LPS given, sublingual intravenous injection of ulinastatin, respectively, after administration 2h, 6h each killed 6 each group, while taking the kidney tissue preparation uniform Pulp and pathological examination. The levels of TNFα, IL6, IL1β and iNOS in serum and kidney tissue were determined by radioimmunoassay. Results The levels of TNFα, IL6, IL1β, iNOS in LPS + ulinastatin group were significantly lower than those in LPS group (P <0.05), and the pathological changes were lighter than those in LPS group. Conclusion Ulinastatin can significantly reduce the levels of TNFα, IL6, IL1β and iNOS in the serum and kidney of MODS rats, and alleviate the renal injury induced by inflammatory cytokines and protect renal function.
