目的研究血虚肝郁证大鼠海马CA1区磷酸二酯酶(PDE)不同亚型蛋白表达的变化及芍药苷的调控作用。方法分为空白组、模型组、芍药苷低剂量组(20 mg/kg)、芍药苷高剂量组(40mg/kg),造模第1天开始给药,空白组、模型组给等量纯水,共造模21 d。治疗结束,灌注固定后取脑组织,石蜡包埋切片,采用免疫组化法检测PDE1A、PDE2A、PDE4A、PDE5A、PDE9A、PDE10A的蛋白表达。结果模型组大鼠海马CA1区的PDE1A、PDE2A、PDE5A、PDE9A蛋白表达显著升高(P<0.05,P<0.01,P<0.01,P<0.01),PDE4A、PDE10A的蛋白表达显著降低(P<0.01,P<0.05),芍药苷可显著下调PDE1A、PDE2A、PDE5A、PDE9A的蛋白表达(P<0.05,P<0.01,P<0.01,P<0.01),明显上调PDE4A、PDE10A的蛋白表达(P<0.05)。结论血虚肝郁证模型大鼠由于受到辐照和束缚双重应激的影响,海马CA1区PDE各亚型表达显著改变,芍药苷能够通过调控不同底物分解酶的表达,从而影响2种核苷酸的含量,表明白芍养血柔肝作用是多靶点多途径的动态平衡。
Objective To study the changes of phosphodiesterase (PDE) subtypes in hippocampal CA1 region of rats with deficiency of blood and liver syndrome and the regulatory effect of paeoniflorin. Methods The rats were divided into blank group, model group, low dose paeoniflorin group (20 mg / kg) and high dose paeoniflorin group (40 mg / kg) Water, a total modeling 21 d. At the end of treatment, brain tissue and paraffin embedded sections were perfusion fixed, and protein expression of PDE1A, PDE2A, PDE4A, PDE5A, PDE9A and PDE10A were detected by immunohistochemistry. Results The protein expressions of PDE1A, PDE2A, PDE5A and PDE9A in CA1 area of hippocampus in model group were significantly increased (P <0.01, P <0.01, P <0.01) (P <0.01, P <0.01, P <0.05). Paeoniflorin significantly decreased the protein expression of PDE1A, PDE2A, PDE5A and PDE9A (P <0.05, <0.05). Conclusion The expression of PDE subtypes in hippocampal CA1 region was significantly changed due to the dual stress of irradiation and tethering in the model of blood deficiency and liver depression syndrome. Paeoniflorin could affect the expression of two substrate enzymes, The content of glycosides showed that the action of nourishing the liver and activating the liver of the white peony is a multi-target multi-channel dynamic balance.
