目的研究不同细胞制备方法对细胞因子诱导的杀伤细胞(CIK)细胞亚群比例的影响。方法将42份CIK细胞随机均分为实验组和对照组。对照组按常规CIK细胞制备方法制备;实验组在对照组基础上于第2天加入重组人粒细胞集落刺激因子(rhG-CSF)10ng/ml。待细胞成熟后采用流式细胞术检测两组CD3+CD4+、CD3+CD8+和CD3+CD56+T淋巴细胞比例的变化。结果与对照组相比,实验组CD3+CD4+T细胞比例下降(P<0.01),CD3+CD56+T细胞比例升高(P<0.05);CD3+CD8+T细胞比例稍高,但无统计学差异(P>0.05)。结论在CIK细胞制备过程中加入rhG-CSF可抑制CD3+CD4+T细胞,增加CD3+CD56+T细胞生成。
Objective To study the effect of different cell preparation methods on the cytokine-induced killer (CIK) cell subsets. Methods 42 CIK cells were randomly divided into experimental group and control group. The control group was prepared according to the conventional CIK cell preparation method. In the experimental group, recombinant human granulocyte colony-stimulating factor (rhG-CSF) 10ng / ml was added on the second day on the basis of the control group. After the cells matured, the proportion of CD3 + CD4 +, CD3 + CD8 + and CD3 + CD56 + T lymphocytes in the two groups were measured by flow cytometry. Results Compared with the control group, the proportion of CD3 + CD4 + T cells in the experimental group decreased (P <0.01) and the proportion of CD3 + CD56 + T cells increased (P <0.05). The proportion of CD3 + CD8 + T cells was slightly higher Statistical difference (P> 0.05). Conclusion The addition of rhG-CSF during the preparation of CIK cells can inhibit CD3 + CD4 + T cells and increase the production of CD3 + CD56 + T cells.
