为探讨IL-6基因转导对人乳腺癌细胞体内外生长特性的影响,我们构建了含人IL-6cDNA的逆转录病毒载体pLIL6SN。采用脂质体介导的方法将pLIL6SN导入MCF-7细胞中,对基因转导细胞的体外生长、粘附特性及裸鼠体内的实验性转移能力进行了测定。实验结果表明,经IL-6基因修饰的MCF-7细胞虽显示了明显的体外生长抑制,裸鼠体内实验性转移能力却获得增强。文中对其可能的影响机制进行了探讨
To investigate the effect of IL-6 gene transduction on the growth characteristics of human breast cancer cells in vitro and in vivo, we constructed retroviral vector pLIL6SN containing human IL-6 cDNA. The liposome-mediated method was used to introduce pLIL6SN into MCF-7 cells. The in vitro growth and adhesion characteristics of the transduced cells and the experimental metastatic ability in nude mice were determined. The experimental results showed that the IL-6 gene-modified MCF-7 cells showed obvious growth inhibition in vitro, but the experimental metastatic ability in nude mice was enhanced. The paper discusses its possible influence mechanism
