本工作的目的系观察肾上腺糖皮质激素对离体培养的人体肝癌细胞(BEL- 7402)引起的形态学表型改变。肝癌细胞经浓度为7.6×10~(-6)的合成糖皮质激素——地塞米松处理48小时显示,体积较对照细胞显著较大,呈扁平多边形。电镜观察到:1)激素处理48小时、浓度为7.6×10~(-7)M及7.5×10~(-6)M的两个剂量组,线粒体偶有增大及融合现象;胞膜下外细胞质区域有短的微管出现。2)处理72小时的两个剂量组,肝癌细胞的微绒毛似有减少;经常见到体积明显增大的线粒体及成片的糖元;胞质内经常出现长的、纵横交错的微管,尤以7.5×10~(-6)M组明显,弥散分布的微丝和张力纤维出现的频率也远高于对照组。本文对肝癌细胞出现的表型变化的意义进行了讨论。
The purpose of this study was to observe the morphological phenotypes induced by adrenocorticotropic hormone on human hepatocellular carcinoma cells (BEL-7402) cultured in vitro. The hepatoma cells treated with synthetic glucocorticoid dexamethasone at a concentration of 7.6 × 10 ~ (-6) for 48 hours showed that the volume of hepatoma cells was significantly larger than that of the control cells and showed a flat polygon. Electron microscopy showed that: 1) Mitochondria were occasionally enlarged and fused under hormone treatment for 48 hours at concentrations of 7.6 × 10 -7 M and 7.5 × 10 -6 M; Short microtubules appear outside the cytoplasmic area. 2) 72h treatment of two dose groups, liver cancer cells seem to reduce the microvilli; often see the volume was significantly increased mitochondria and into pieces of glycogen; cytoplasm often appear long, criss-crossing the microtubules, Especially in 7.5 × 10 ~ (-6) M group, the frequency of occurrence of microfilament and tension fiber with diffuse distribution was also much higher than that of the control group. This article discusses the significance of phenotypic changes in hepatocellular carcinoma cells.
