目的建立龙胆药材的指纹图谱研究方法并进行聚类分析。方法色谱柱为Shimadzu C18(3.0mm×75mm,1.7μm);流动相:乙腈-0.4%磷酸(5.5∶94.5),体积流量:0.8mL/min;检测波长:240nm;柱温为40℃。结果獐牙菜苦苷、龙胆苦苷分别在0.1548~1.858mg/mL,0.322~5.152mg/mL呈良好的线性关系,平均回收率分别为102.09%,RSD=1.68%(n=6);103.80%,RSD=1.11%(n=6)。指纹图谱的精密度与重现性试验中的各色谱峰的相对保留时间与相对峰面积的RSD小于2.1%,指纹图谱相似度结果与聚类分析谱系图一致。结论建立的指纹图谱适用于龙胆药材的标准化种植及质量控制。
OBJECTIVE To establish the research methods of fingerprinting gentian herbs and to conduct cluster analysis. Methods The column was Shimadzu C18 (3.0mm × 75mm, 1.7μm). The mobile phase consisted of acetonitrile-0.4% phosphoric acid (5.5:94.5), volume flow rate 0.8mL / min, detection wavelength 240nm and column temperature 40 ℃. Results Swertia glycosides and gentiopicroside had good linearity in the ranges of 0.1548 ~ 1.858mg / mL and 0.322 ~ 5.152mg / mL, with the average recovery of 102.09% and RSD = 1.68% (n = 6), respectively. 103.80%, RSD = 1.11% (n = 6). The RSDs of relative retention time and relative peak area of each chromatographic peak in the precision and reproducibility test of fingerprints were less than 2.1%, and the similarity of fingerprints was consistent with that of the cluster analysis. Conclusion The established fingerprinting is suitable for the standard planting and quality control of gentian medicine.
