目的 探讨硒、碘对培养大鼠海马神经细胞原癌基因c fos、c jun表达的影响。方法 以无血清培养大鼠海马神经细胞为实验对象 ,培养液中加入不同浓度的碘、硒 ,用盖玻片培养法 ,在培养的 1、3、5、7和 10d时对海马神经细胞进行c fos、c jun的mRNA原位分子杂交和蛋白产物的免疫组织化学SABC法染色 ,对杂交信号和免疫阳性产物进行计算机图像分析。结果 硒可明显促进海马神经细胞原癌基因c fos、c junmRNA及其蛋白的表达。硒、碘合用的促进作用最明显 ,尤其是c junmRNA表达。结论 碘和硒可通过促进海马神经细胞即刻早期基因c fos及c jun表达 ,特别是c jun表达 ,进而影响神经功能发育
Objective To investigate the effects of selenium and iodine on the expression of proto-oncogene c-fos and c jun in cultured hippocampal neurons. Methods Rat hippocampal neurons were cultured in serum-free medium with different concentrations of iodine and selenium. Cultured hippocampal neurons were cultured at 1, 3, 5, 7 and 10 days c fos, c jun mRNA in situ hybridization and protein products by immunohistochemical SABC staining hybridization signal and immunopositive products for computer image analysis. Results Selenium could obviously promote the expression of proto-oncogene c fos, c jun mRNA and protein in hippocampal neurons. The promotion effect of selenium and iodine combination is the most obvious, especially c junmRNA expression. Conclusion Iodine and selenium may affect the development of neural function by promoting the expression of immediate early genes c fos and c jun, especially c jun, in hippocampal neurons
