目的建立HL-7702细胞BMP7基因转染细胞系,并检测其增殖和迁移能力的变化。方法脂质体介导方法转染细胞,采用RT-PCR、western-blot方法检测其mRNA及蛋白表达以评价其转染效果;用MTT、划痕实验检测转染后增殖及迁移能力的变化。结果成功建立转染细胞系,转染BMP7基因后HL-7702细胞增殖和迁移能力有所增强。结论 BMP7基因高表达可能是肝细胞癌变的原因之一。
OBJECTIVE: To establish a cell line transfected with BMP7 gene of HL-7702 cells and detect their proliferation and migration abilities. Methods Lipofectamine was used to transfect cells. The expression of mRNA and protein was detected by RT-PCR and western-blot to evaluate the transfection efficiency. The proliferation and migration ability of the transfected cells were detected by MTT assay and scratch assay. Results The transfected cell lines were successfully established. The ability of proliferation and migration of HL-7702 cells after transfection with BMP7 gene was enhanced. Conclusion The high expression of BMP7 gene may be one of the reasons of hepatocellular carcinoma.
