目的通过荧光光漂白恢复技术观察PC12细胞在纳米细菌纤维素上的生长作用。方法制备纳米细菌纤维素膜并与PC12细胞共培养,通过FRAP技术利用激光共聚焦观察PC12细胞间缝隙连接的通讯功能。结果细胞内荧光被淬灭后可通过缝隙连接通道得到一定程度的恢复,在胞内荧光漂白后第4分钟时,实验组的平均荧光漂白恢复率为(22.3±0.39)%,与对照组的(21.8±0.85)%相比差异无统计学意义。结论 PC12细胞可在细菌纤维素膜上良好生长,能维持正常的细胞间通讯功能。
Objective To observe the growth of PC12 cells on nanobacterial cellulose by fluorescence photobleaching. Methods Nano-bacterial cellulose membranes were prepared and co-cultured with PC12 cells. The communication function of PC12 cell gap junctions was observed by laser scanning confocal microscopy (FRAP). Results After fluorescence quenching, the intracellular fluorescence was restored to a certain extent through the gap junction channel. At 4 minutes after intracellular fluorescence bleaching, the mean fluorescence recovery of the experimental group was (22.3 ± 0.39)%, which was significantly lower than that of the control group (21.8 ± 0.85)% compared with the difference was not statistically significant. Conclusion PC12 cells can grow well on bacterial cellulose membranes and maintain normal cell-cell communication.
