目的检测microRNA-150(miR-150)在结肠癌细胞株、结直肠癌(CRC)组织中的表达,分析其与临床特征间的关系。方法采用实时荧光定量逆转录聚合酶链反应(qRT-PCR)检测8种结肠癌细胞株及45例CRC患者癌组织及对应正常组织中miR-150的表达情况,分析其表达与CRC临床病理资料之间的关系。结果 miR-150在8种结肠癌细胞株中表达明显降低,CRC组织中miR-150的表达较同一患者正常黏膜组织中的miR-150表达明显下调(P<0.05);miR-150在CRC组织中的表达与肿瘤的分化程度(P<0.05)及病理类型(P<0.01)密切相关。结论 miR-150在CRC中表达下调,其低表达与CRC的分化及病理类型相关,可作为结直肠癌诊断及治疗的指标之一。
Objective To detect the expression of microRNA-150 (miR-150) in colon cancer cell lines and colorectal cancer (CRC) tissues and to analyze its relationship with clinical features. Methods The expression of miR-150 in 8 kinds of colon cancer cell lines and 45 cases of CRC patients and their corresponding normal tissues were detected by qRT-PCR. The expression of miR-150 was analyzed with the clinicopathological data of CRC The relationship between. Results The expression of miR-150 in 8 colon cancer cell lines was significantly decreased. The expression of miR-150 in CRC tissues was significantly lower than that in normal mucosa (P <0.05). The expression of miR-150 in CRC tissues (P <0.05) and pathological type (P <0.01). Conclusion The expression of miR-150 is down-regulated in CRC. The low expression of miR-150 is associated with the differentiation of CRC and the pathological type, which may be used as one of the indicators for the diagnosis and treatment of colorectal cancer.
