使用高效液相色谱法分离纯化了小链霉菌HCCB10043的重要代谢产物,获得3个精制品;经高分辨质谱、二级质谱与氨基酸分析鉴定其分别为A21978C1、C2和C3。菌株16S rDNA与已知的A21978C产生菌玫瑰孢链霉菌NRRL11379的同源性为99.2%,系统发育分析显示两者的亲缘关系很近;经基因钓取与沉默验证,结果显示该菌株的A21978C生物合成基因簇与玫瑰孢链霉菌报道完全相同。
High performance liquid chromatography (HPLC) was used to separate and purify the important metabolites of HCCB10043 and obtain 3 purified products. A21978C1, C2 and C3 were identified by high-resolution mass spectrometry, secondary mass spectrometry and amino acid analysis. The homology of the 16S rDNA of the strain with the known strain A21978C producing Streptomyces roseus NRRL11379 was 99.2%. Phylogenetic analysis showed that the 16S rDNA was closely related to each other. The results of gene-based fetch and silencing showed that the A21978C The synthetic gene cluster is identical to the one reported by Streptomyces roseosporus.
