目的探讨雌激素对大鼠脂肪源干细胞(adipose-derived stem cells,ADSCs)增殖及Ⅰ、Ⅲ型胶原蛋白表达的影响。方法 2016年12月至2017年5月以广州医科大学第一附属医院保存的大鼠ADSCs作为研究对象,体外培养过程中加入不同浓度17-β雌二醇,MTT检测增殖能力,荧光定量PCR及蛋白质印记技术(Western blot)检测Ⅰ、Ⅲ型胶原蛋白mRNA表达及蛋白的表达情况。结果 17-β雌二醇浓度为10-9mol/L时,大鼠ADSCs增殖指数及Ⅰ、Ⅲ型胶原蛋白的表达明显增加,差异有统计学意义(P<0.05)。结论 17-β雌二醇可以促进ADSCs胶原蛋白的表达,两者联合应用有望成为治疗绝经后盆腔器官脱垂的方法之一。
Objective To investigate the effect of estrogen on the proliferation and the expression of collagen type Ⅰ and Ⅲ in adipose-derived stem cells (ADSCs) of rats. Methods From December 2016 to May 2017, ADSCs of rats preserved in the First Affiliated Hospital of Guangzhou Medical University were used as research objects. Different concentrations of 17-β-estradiol were added during the culture in vitro. MTT assay was used to detect the proliferative capacity. Western blot was used to detect the mRNA expression of type I and type III collagen and protein expression. Results When the concentration of 17-β-estradiol was 10-9mol / L, the proliferation index of ADSCs and the expression of type I and type III collagen were significantly increased (P <0.05). Conclusion 17-β-estradiol can promote the expression of collagen in ADSCs, and the combination of them may be one of the methods to treat post-menopausal pelvic organ prolapse.
