目的 研制抗组织型纤溶酶原激活物 (tPA)单克隆抗体 (mAb) ,鉴定其各种生物学特性。方法 采用传统的细胞融合、有限稀释法 ,制备稳定分泌mAb的细胞株。采用辛酸、硫酸铵沉淀法 ,纯化腹水中的mAb ,以SDS -PAGE鉴定其纯度。运用ELISA和细胞免疫组化染色 ,鉴定mAb的各项特性 .结果 获得了两株能稳定分泌抗tPAmAb的细胞株。用该两株细胞分泌的mAb能检测出 0 .4 μg/L重组的tPA和血清中的tPA。结论 成功地研制了两株抗tPA的mAb ,为进一步用于临床诊断和实验研究创造了条件
Objective To develop a monoclonal antibody (mAb) against tissue plasminogen activator (tPA) and identify its various biological characteristics. Methods The cell lines stably secreting mAbs were prepared by the traditional cell fusion and limited dilution method. Using octanoic acid, ammonium sulfate precipitation, purification of ascites mAb, SDS-PAGE to identify its purity. The characteristics of mAb were identified by ELISA and immunohistochemical staining.Results Two cell lines stably secreting anti-tPA mAb were obtained. The mAbs secreted by these two cells detected 0. 4 μg / L recombinant tPA and serum tPA. Conclusion Two mAbs against tPA were successfully developed, which provided conditions for further clinical diagnosis and experimental study
