目的探索外源性硫化氢对THP-1源性巨噬细胞脂质蓄积的影响及机制。方法以硫氢化钠(NaHS)做为外源性硫化氢的供体,采用油红O染色结合光密度法测细胞内脂质含量,RT-PCR和Westernblot法测SR-A mRNA和蛋白表达。结果氧化型低密度脂蛋白(ox-LDL)显著诱导巨噬细胞内脂质蓄积和SR-A表达,而NaHS呈浓度依赖性降低ox-LDL诱导的细胞内脂质蓄积,并显著下调SR-A mRNA和蛋白水平。结论外源性硫化氢可能通过抑制SR-A表达减少ox-LDL诱导的THP-1源性巨噬细胞脂质蓄积。
Objective To explore the effect and mechanism of exogenous hydrogen sulfide on lipid accumulation in THP-1-derived macrophages. Methods Sodium hydrogen sulfite (NaHS) was used as donor of exogenous hydrogen sulfide. The intracellular lipid content was determined by oil red O staining and optical density. The mRNA and protein expression of SR-A were detected by RT-PCR and Western blotting. Results Ox-LDL significantly induced lipid accumulation and SR-A expression in macrophages, while NaHS decreased ox-LDL-induced intracellular lipid accumulation in a concentration-dependent manner and significantly down-regulated the expression of SR- A mRNA and protein levels. Conclusion Exogenous hydrogen sulfide may reduce ox-LDL-induced lipid accumulation in THP-1-derived macrophages by inhibiting SR-A expression.
